ACCESSING HIDDEN THERAPEUTICS IN NATURE
Silenyx is a Princeton-based biopharmaceutical company that is opening new doors to drug discovery by tapping into hidden therapeutics in nature. Although historically prolific, recent investments into natural products have delivered diminishing returns due to the costly rediscovery of known compounds. This is in part due to the silent nature of most biosynthetic genes: conventional methods only capture ~10% of a bacterium’s biosynthetic potential. Using a broadly applicable, high-throughput, genetics-free, and cloning-free approach, Silenyx provides the simplest and fastest natural product drug discovery platform which can rapidly activate silent biosynthetic genes, unlocking the full potential of any culturable microbe.
Our Platform, HiTES (High‐Throughput Elicitor Screening) is a facile, rapid, patent-protected and high-throughput method for activation of any desired silent biosynthetic gene cluster. Recent research has shown that 80‐90% of all biosynthetic gene clusters – the sets of genes responsible for the synthesis of a given natural product – are ‘silent’ or ‘cryptic’, meaning their small molecule products are not biosynthesized under standard growth conditions. Accordingly, these cryptic natural products have not been interrogated in routine drug discovery screens.
